circPDE4B Regulates the ECM Degradation and Apoptosis of Chondrocyte Through miR-7 in Osteoarthritis

Abstract:Objective To investigate the effect and mechanism of circPDE4B regulating miR-7 on the extracellular matrix (ECM) degradation and apoptosis of chondrocyte in osteoarthritis(OA).Methods RT-qPCR was used to detect the expression levels of circPDE4B and miR-7 in OA cartilage tissues and chondrocytes.After OA chondrocytes were transfected with oe-NC and oe-circPDE4B,or si-NC,si-circPDE4B and co-transfected with si-circPDE4B+miR-7 inhibitors,the expression levels of ECM related molecules SOX9,COL2A1,MMP3 and MMP13 were detected by RT-qPCR and Western Blot;and flow cytometry was used to detect cell apoptosis.The dual luciferase reporter gene was used to detect the binding of circPDE4B and miR-7.Results The expression level of circPDE4B in OA cartilage tissues and chondrocytes was significantly lower than that of normal cartilage tissues and chondrocytes (P<0.05).Overexpression of circPDE4B significantly inhibited the apoptosis of OA chondrocytes and the expression level of apoptosis-related molecule Cleaved-caspase-3 (P<0.05).Meanwhile,overexpression of circPDE4B significantly promoted the expression levels of SOX9 and COL2A1 (P<0.05),and inhibited the expression levels of MMP3 and MMP13 (P<0.05).The results of the luciferase reporter gene test showed that circPDE4B could target miR-7 (P<0.05),and overexpression of circPDE4B inhibited the expression of miR-7 (P<0.05).The expression levels of circPDE4B and miR-7 in cartilage tissues of patients with OA were negatively correlated (P<0.05).Co-transfection of si-circPDE4B and miR-7 inhibitors reversed the accelerating effect of single transfection of si-circPDE4B on OA chondrocyte apoptosis and ECM degradation (P>0.05).Conclusion circPDE4B is significantly low expressed in OA cartilage tissues and chondrocytes.CircPDE4B inhibites OA chondrocyte apoptosis and cartilage ECM degradation by targeting miR-7.
