Abstract Objective To study the animal model of sciatic nerve injury by combining poly[(lactic-co-glycolic acid),PLGA]catheters with bone marrow mesenchymal stem cells (bone mesenchymal stem cells,BMSCs) combined with extracellular matrix (extracellular matrix,ECM),biomechanical properties of the sciatic nerve after gel transplantation and provide basic research support for clinical application.Methods The rat sciatic nerve injury model was transplanted with autologous nerve,PLGA catheter combined with BMSCs,PLGA catheter combined with BMSCs and ECM gel for 6,12,18,and 24 weeks,and the general behavior of rats in each group was observed.The functional index of the sciatic nerve was measured 168 days after the transplantation in the rats,and then the sciatic nerves of the rats in each group were taken for tensile mechanical properties experiments.Results The tensile elastic limit load,elastic limit stress,maximum load,maximum stress,maximum strain,and elastic limit strain of the PLGA catheter combined with BMSCs transplantation group and the PLGA catheter combined with BMSCs and ECM gel transplantation group were greater than those of the autologous nerve transplantation group.The difference was statistically significant (P<0.05).The tensile elastic limit load,elastic limit stress,maximum load,maximum stress,and elastic limit strain of the PLGA catheter composite BMSCs and ECM gel transplantation group were greater than those of the PLGA catheter composite BMSCs transplantation group.The difference was statistically significant (P<0.05) Conclusion PLGA catheter combined with BMSCs,PLGA catheter combined with BMSCs and ECM gel transplantation have obvious repair effect on sciatic nerve injury.
