Evaluation of Compatibility,Osteogenesis And Antibacterial Activity of Zn-2Ag-0.4Mg Scaffolds In Vitro

Abstract:Objective To prepare a Zn-2Ag-0.4Mg porous alloy scaffold and study its compatibility,osteogenesis and antibacterial properties in vitro.Methods In this study,pure silver and magnesium were fused into Zn-2Ag-0.4Mg porous alloy under the protection of Ar+2%SF-6 at 650℃.The alloy was divided into zinc group,zinc silver group,zinc silver magnesium group,and control group.The cytotoxicity was measured by Methyl Thiazolyl Tetrazolium (MTT) method.The cytoskeleton was stained with penicin method,the osteogenic properties were tested by alizarin red staining and alkaline phosphatase (ALP) measurement,and the antibacterial properties were tested by the mode of Staphylococcus aureus co-culture.Results Zinc,magnesium and silver alloy had lower cytotoxicity.The experimental results further confirmed that zinc-magnesium-silver alloy had lower toxicity,and thus had better cytocompatibility,better osteogenesis and antibacterial properties.Conclusion The addition of magnesium improves the osteogenic properties of the material,the addition of silver significantly improves the antibacterial properties of the material,and the addition of two elements improves the cytocompatibility and osteogenic properties of pure zinc.Therefore,zinc magnesium silver alloy is expected to be a new choice material for orthopedic implants,especially in the field of bone defect or fracture repair.
