Abstract:Objective To study the conditions of inducing C57BL/6 mouse bone marrow cells to osteoclasts in vitro before and after purification,and summarize the techniques of inducing osteoclasts by primary cells.Methods Weinoculated the primary cells before and after purification with different densities (ρ=5×105 cells/cm2 and ρ=2.5×105 cells/cm2) and added the induction factors at different stages to observe the number of tartrate-resistant acid phosphatase (TRAP) staining positive cells and the expression level of osteoclast marker genes in each group.Results Unpurified bonemarrowcells with low density inoculation (2.5×105 cells/cm2) could not be induced into osteoclasts.After increasing inoculation density (5×105 cells/cm2),osteoclasts couldbe induced by co-stimulation of M-CSF and RANKL.Purified cells inoculated with low density (2.5×105 cells/cm2) couldbe induced into osteoclasts whatever by either M-CSF pretreatment or co-stimulation with RANKL,and the induction effect of the M-CSF pretreatment group was better.Conclusion When using primary bone marrow cells to induce osteoclasts formation and the proportion of mononuclear cells was low,priority should be given to increasing the cell inoculation density and adding RANKL as early as possible to promote the differentiation of mononuclear cells into osteoclasts.
